How to scale a scientific chart to read its data

Dear All,

I need to read data from a chart that was published in a scientific journal.Maybe it 's made originally in Excel but now its a picture in my hands.
I open it in Photoshop or AutoCAD and show grids.
Please tell me two things:

1-How I can set the point (x=y=0) manually in left down corner of my chart?

2-Is there any way I can tell the software that length between point A and point B (on my chart) is equal to 1cm.
OK photoshop or AutoCAD?
Now regarding this scale that I introduced to you right now, tell me the coordinate of any point I click or length between two other points that I want.
Also if you know any way I can write a simple application with java to do it for me please tell me some guidelines.

I greatly aknowledge any help or suggestion.If there is any software or way you know that can help me it's again appreciated to inform me.
