where and how to specify access specification

hai
my doubt is where should we specify access specification and how
actually there will be one person who can have access to some area only
and other person can access to some other area.
for this i think, i need to specify access specification at deployment discriptor using getCallerPrincipal() or is CallerInrole() from context interface if it is for servlets.
is it correct or is there any other mean to do this
And one more thing if i want to specify access to 100 members. shell i can do it in DD itself for all 100 members or any other mean is there.
