I would like to upgrade my Websphere Application Server5.0 to Websphere Application Server6.0.Do you know the procedure please let me know

It's not a smooth upgrade....I mean it's not a fixpack to apply on your existing WAS v5.x to make it WAS v6.x..
1. Get new WAS v6.x software from your procurement
2. Blow off your existing WAS v5.x and then
3. Install WAS v6.x from scratch.
